HLA-A Low Resolution Typing

Clinical information

Diagnostic Utility:

HLA typing at low resolution allows us to determine HLA typing at the allelic group level (2 digits) for:

  • Family members of patients candidates for hematopoietic progenitor cell transplantation.
  • Confirmation of HLA typing for patients candidates for hematopoietic progenitor cell transplantation and their donors (both related and unrelated).
  • Patients candidates for solid organ transplantation.
  • Verification of units of umbilical cord blood.
  • Study of the maternal sample of umbilical cord blood units (maternal haplotype).

Method:

PCR-SSO for the HLA-A locus.

Exons 2 and 3 of the HLA-A gene are amplified by PCR using biotinylated primers. The amplified products are hybridized with specific oligonucleotide probes immobilised on microspheres. Subsequently, the addition of streptavidin-PE buffer allows visualisation of the hybridization pattern using the LUMINEX system.

Reference Values

With low-resolution typing, two results are obtained for each HLA gene, for example: HLA-A*02 A*26.
