Diagnostic Utility:
HLA typing by low resolution allows us to determine the HLA typing at the allelic group level (2 digits) for:
- Relatives of patients who are candidates for hematopoietic stem cell transplantation.
- Confirmation of HLA typing for patients who are candidates for hematopoietic stem cell transplantation and their donors (both related and unrelated).
- Patients who are candidates for solid organ transplantation.
- Verification of umbilical cord blood units.
- Study of maternal samples of umbilical cord blood units (maternal haplotype).
Method:
PCR-SSO for HLA-DQB1 and HLA-DQA1 loci.
Exons 2 and 3 of the HLA-DQB1 and HLA-DQA1 gene are amplified by PCR using biotinylated primers. The amplified products are hybridized with specific oligonucleotide probes immobilized on microspheres. Subsequently, the addition of streptavidin-PE buffer allows visualization of the hybridization pattern using the LUMINEX device.
Reference Values
With low-resolution typing, two results are obtained for each HLA gene, for example: HLA-DQB1*02 DQB1*03, DQA1*05 DQA1*03.
